Overview
A novel series of dual-targeted bifunctional anti-flu drugs comprising of influenza virus inhibition and pro-inflammatory cytokine suppression are designed, synthesized and evaluated. In vitro and in vivo functional analyses have shown remarkable protection of cells and mice by oral administration against H1N1 & H5N1 influenza viruses.

Influenza is an infectious disease caused by influenza viruses. The type A viruses, further subdivided into several serotypes, are the most virulent human pathogens among the three influenza types and cause the severest clinical symptoms. The outbreaks of H5N1 in 1996 and H1N1 in 2009 not only caused pandemic deaths but also revealed the problems of drug-resistant viral strains.  Patients of H5N1 frequently associated with severe inflammation and do not response well to the two major anti-flu drugs, Relenza and Tamiflu。
Moreover, greater than 90% of Type A viruses have become resistant to Relenza and Tamiflu.  The anti-flu drugs on the market are not sufficient to protect us from pandemic flu. The development of new anti-flu drugs with both anti-viral and anti-inflammatory functions is an important clinical unmet need and has great market potential.
